Many people say that a completely dead room will take away from how the vocals sound and this is absolutely true. You want your booth to be soundproof so no air gets in or out but you also have to take proper care to do it right or your vocals could come out sounding butchered. There will be no need for measurements and everything like that unless you're trying to make a professional vocal booth with top of the line capabilities and options. To see what kind of space you're working with you have to clean out the entire closet so it's basically empty. After cleaning it out evaluate it by talking in each one of the corners, talk LOUD. You figure it like this; if you wanted echo effects you could always add them into your mix with a basic plug in on your audio program or even a analog effects generator. Just as a quick fact, many people set up their vocal booths in bathrooms as it gives them the naturally sounding reverb once the sound bounces off of the tiles. But since we're dealing with the closet, we want it just about dead but not completely. Although the size of the closet isn't really important, have common sense when building. Make sure you can turn around in the closet and you actually have enough room for comfort. It's nothing worst than being in a vocal booth that's either to small or too hot, believe they heat up extra fast.
