One can only guess what will happen in the next go around for Avatar. James Cameron will likely capitalize on the new movie making techniques developed in the first Avatar movie.
He should also look to improve the story line. If there is anything to criticize about the first Avatar movie, it would be the story line. The plot the first Avatar movie was is a blend of many themes that have been covered in Hollywood. Most glaringly themes covered by the 1990 hit Dances with Wolves. Avatar was even parodied in by South Park in there now infamous Dances with Smurfs episode in November, 2009.

Forecasting exactly where the story of Avatar will lead in the sequel is a difficult proposition. The most plausible jump off point will to start at the ending point of the first Avatar. Jake awakens after having been restored by the transfer of his conscience through the Tree of Souls. He will still need to make some adjustments to his new life. A family may be in the offing with his new mate. Also, some humans remained behind, so there may be some back story about them.
Pandora is not the only moon orbiting the large gas giant planet that was so prominently displayed in the Pandoran skyline throughout the movie. Perhaps there are other sentient beings on the other moons or planets that the N'avi will be able to contact. If the humans left behind any technology, there may be some debate amongst the N'avi as to how to make use of it. Will they want to make use of the technology, and will it create a whole new set of problems for the N'avi. All these themes are fairly standard plot lines that Cameron can follow for the sequel to Avatar.
Pandora was a natural resource treasure house for Earthlings. The first Avatar movie dealt with the greed of humans. Cameron drew interesting parallels about how the consequences of our actions today will effect Earth in the future. The sequel to Avatar will likely try open up a new spiritual front on how humans on Earth view Pandora. Word could get back to Earth about Jake's survival under the Tree of Souls, opening the door to immortality for humans.
Avatar portrayed the N'avi attempting to save two lives under the Tree of Souls during the movie. One attempt fails and the character played by Sigourney Weaver, Dr. Augustine dies. The other attempt is successful with the aforementioned transfer of Jake's conscience.
What if word of of the Tree of Souls reached Earth in the sequel for Avatar? Will rich Earthlings start spending huge sums of money to create their genetically matched human or N'avi to get a new lease on life. Will Eywa allow it? Or will she rebel once again? If humans are able to understand the genetic code of the trees and their network, perhaps they can perfect a virus to bend the Pandoran biological network to do their bidding.
Do the N'avi hold darker secrets. Once they realize that they can transfer consciences, will N'avi seek immortality by creating their own Avatars? Perhaps a power hungry and sinister N'avi will rise up and attempt to usurp control of the territory surrounding the Tree of Souls.
Do not expect is a rehash of the old storyline, with humans coming back with a bigger and larger force to try to take the planet over from the N'avi. The quest for unobtainium is over. James Cameron and his crew are sure to come up with something new and imaginative to take movie theaters by storm once again.
