Four years after the incidents that happened in the second film, Doctor Alan Grant is approached by a supposedly wealthy couple, Paul and Amanda Kirby, who want to hire him to take them on a tour of the island of Isla Sorna. He is very reluctant to do this until he is offered by Paul Kirby to fund his dig. He figures something is up when the plane that he, his protege, Billy Brennan, the Kirby's and some mercenaries, land on the island. They are attacked by a huge dinosaur when Amanda Kirby starts yelling for their son. They attempt to escape in the plane but they run into the huge dinosaur and crash. The crash results in two deaths and the survivors flee into the jungle, trying to escape the huge dinosaur. They run into a T-Rex, which attempts to kill them until it runs into the huge dinosaur which we learn is called a Spinosaurus. Dr. Grant discovers the real reason they are on the island and he is not happy. He makes the decision to lead them to the coast so they can get off the island, despite the protests. Along the way, they do discover a dead body and the fact that the Kirby's son is alive. They also discover a Velociraptor nests and unknown to Dr. Grant, Billy manages to take two, which results in them being hunted and attacked by the Velociraptors.
Dr. Grant manages to get separated from the group and ends up finding the Kirby's son, Eric, who has managed to survive the eight weeks that he has been on the island. They end up reuniting with the Kirby's and Billy and are again attacked by the Spinosaurus. They manage to find shelter where Dr. Grant learns about the egg that Billy took. He is angry and upset with Billy and they have words. They leave the shelter to make their way down to the river where they have to enter a huge aviary where they are attacked by numerous Pteranondons. Eric is taken by the adults and Billy tries to rescue him but is attacked by the adults. Thinking he is dead, Dr. Grant and the others decide to leave on the boat that they find. They are again attacked on the river by the Spinosaurus and Dr. Grant manages to contact Ellie to call for help.
They are almost to freedom where they can find a rescue when they are surrounded by the Velociraptors that had attacked them earlier. They want their eggs back and they want Ellie to do the deed. Dr. Grant almost manages to get them killed when he tries to imitate their call with a raptor head, but some noises distract them and they leave with the eggs. They hear a helicopter overhead and run to the beach where they see a man and then the Marines landing to rescue them. After they board the helicopter, they find out that Billy is alive and Grant is relieved. As the helicopter leaves the island, they see several Pteranondons flying past them, heading away from the island. They wonder where they are heading but joke around about where they are heading. Amanda makes a reference to their home town and that they better not be heading there as they watch them fly off.
