Gregory House (played by Hugh Laurie) is the Head of a Diagnostic Medicine department in a major hospital. The job of Dr. House and his team is to diagnose patients whose problems can't be solved by other departments in the hospital. Usually Gregory House saves the patient but not always which gives the show some realism.
British actor Huge Laurie does an astonishingly good job as Dr. Gregory House. Lisa Edelstein plays Dr. Lisa Cuddy who runs the hospital and is Greg House's love interest after years of teasing that in the plot.
Robert Sean Leonard plays Dr. James Wilson who is "House's" best friend and former roommate. Omar Epps plays Dr. Eric Forman (and used to play on "ER") and at one time left the team and then later returned.
Jessie Spencer plays Dr. Robert Chase an Australian Dr. formerly on House's team and the husband (now separated) of Dr. Allison Cameron played by Jennifer Morrison. Cameron like Chase is a former member of House's Diagnostic Team but remains on the show.
Gregory House can be an ass to his friends and to patients but is great doctor. "House" is also a drug addict currently clean. He had become addicted to Vicoden after he suffered muscle death in his leg, giving him a limp and a cane.
The much anticipated House/Cuddy romance was even faked once when it appeared to viewers that they had "Hooked up." But that romance turned out to be delusions by "House" due to drugs and mental illness which ended him up in treatment and almost costing him his job. Currently in the 2010 season he and Cuddy are a real couple after she dumped her fiancée.
Dr. House can be abusive to his friends and especially his team but occasionally shows a human side. In the "Fetal Position" episode in 2007 "House" wanted to abort a baby to save the mother. When the mother refuses the team decides to remove the baby from the womb to treat it, and then return it. During this procedure the unborn baby reaches out from the womb and grabs "House's" finger. "House" was able to save both mother and child in what was one of the most moving episodes of any television show I have ever watched.

House used to be one of my favs but I don't watch it anymore:( Hugh Laurie was great in the English comedy Black Adder which is hilarious & entertaining for anyone who likes history:)
I used to love this show, but now he's getting on my nerves, lol!
I don't watch TV, but my kids have seen this and think it's good.

I like Hugh Laurie, but only watch occasionally. You summed it up pretty well for anyone new to the show.
