Stop Look and Listen! These three little words will get you very far when trying to locate the Poe Ghosts. Remember that Poe's can only be seen and destroyed when Link is transformed into a Wolf and only if you use your senses to see them. Keep this important fact in mind when roaming the fields at night.
Stop: So you know that in the first part of this quest you're going to have to find at least 20 Poe Souls, and later on you'll be required to find more then that, so judging by the map of Hyrule, there is not that many places a Poe can hide for very long. While you have some spare time, or are stuck trying to figure out what to do next (which may happen more often then you think) take a moment to go into first person view and look around. Doing this will give you, the player, a 360 degree view of the area. You don't have to be in Wolf form to do this either, because a Poe's Lantern will show up with or without your senses active, but going first person in Wolf form with your senses active might also be a god idea.
Look: Make sure to explore areas that may be suspicious. Check areas that are out of the way on your map, or that seemingly have no other purpose in the game. One key tip in not only Zelda, but in any video game is that there is rarely an area that serves no purpose. You may have to light torches, move a block, set a bomb, whatever, but there are purposes to most areas, and a Poe ghost may just be the thing you're looking for in any specific area. Also make sure you look all around areas as well. Don't just glance and be done with it, take time to check behind rocks, down streams, in trees, behind buildings, and so on.
Listen: This aspect of your search for Poe Ghosts will actually be one of the most useful in ways of detecting a Poe. Poe Ghosts carry lanterns with them at all times and in doing so the lanterns jingle when the Poe Ghost moves. It's not a sound that is easily lost to other noises in the game, and is very distinctive. This simple yet crucial tip of listening to your surroundings will allow you to detect Poe that may otherwise have escaped you. Listening for the tinkling of the Poe's lantern will let you know if a Poe is hiding behind a wall, statue, hovering above, or even in the next room.
The quest for finding the Poe Ghosts may seem daunting and irritating at times, but just think of the rewards that lie ahead, and to ease the frustration follow the above tips to find those elusive Poe Ghosts.
